Tepezza (teprotumumab-trbw) was approved by the FDA in January 2020 as the first drug to treat thyroid eye disease (TED). Manufactured by Horizon Therapeutics — now owned by Amgen following a $27.8 billion acquisition — Tepezza was aggressively marketed as a breakthrough therapy.
What patients weren't told: Tepezza carried a serious risk of permanent hearing damage. Horizon's original label suggested roughly 10% of patients might experience hearing-related side effects. Independent research told a very different story — a 2021 study presented to the Endocrine Society found that as many as 65% of patients experienced hearing symptoms, including hearing loss, tinnitus, and ear fullness.
It wasn't until July 2023 — more than three years after launch — that the FDA required Horizon to add a warning for severe and potentially permanent hearing impairment to Tepezza's label. Lawsuits filed nationwide allege Horizon knew about this risk far earlier and failed to warn patients and physicians.
